Spectrophotometric Determination of Aminoglutethimide by Direct, First and Second Derivatives Methods

Abstract:
Spectrophotometric methods are described for the determination of aminoglutethimide in pure samples as well as in formulations. Making use of the appearance of the absorption band at about 242 nm and a small shoulder at about 282 nm in neutral methanol. The direct, first and second derivative spectrophotometric procedures are suggested for the determination of about 0.4 - 20 μg/ml of aminoglutethimide. The effect of the different pH on the absorption band is discussed. The method has been successfully applied to the analysis of the pharmaceutical formulations. The results have been statistically compared with those obtained by the compendial (diazotization) method. Procedural details and data for the three methods are presented. It was proved that the proposed first derivative method can be used for the determination of aminoglutethimide in biological fluids without interferences from its major metabolite N-acetyl aminoglutethimide.
