Hyperglycemic Effect of Calcium Antagonists Related Serum Chromium Level in Rats

ABSTRACT:

Investigations on the effect of calcium antagonists, nifedipine and diliiazem on serum glucm insulin and chromium levels nave been carried out in tne presence and absence of oral antidiabetic agents in rats. Results show that nifedipine (10 mg/kg i.p.) or diliiazem (20 mg/kg orafa caused marked hyperglycaemia associated with significant reduction in serum chromium level Diltiazem was found to decrease serum insulin ievel while nifedipine exerted no effect. DiUiazem-induced hyperglycacmia was counteracted by concurrent administration of glibenclamide *5 n*. kg orally, metformin (500 mg/kg orally) or their combination. Nifedipine induced hyperglycemia in rats was counteracted by concurrent administration of metformin (500 mg/kg P.O.). However, glibenclamide (5 mk/kg P.O.) did not change the hyperglycaemic effect of nifedipine. Adeno¬sine, a hyperglycaemic agent might be implicated in nifedipme-induced hyperglycemia since nifedipine decreases adenosine uptake by tissues. It is concluded that nifedipine-or dilnazem-induced hyperglycaemia might be related to its effect on serum chromium level. The effect of nifedipine on adenosine uptake is also considered as a possible factor.
