Influence of Tetracyline and Penicillin-G-Sodium on the Synthesis in Lipoxygenase Product-( 12-Hete) by the Rat Trachea and Testis

The influence of treatment of rats with tetracycline hydrochloride and penicillin-G sodium on 12-hydroxy eicosa tetraenoic acid (12-HETE) synthesis by the rat testis and trachea was examined. 12-HETE was isolated and quantified using TLC and spectrophotometry at 239 nm. Tetracycline (62.5-250 mg kg-1 day-1 i. p. for 4 days) stimulated 12-HETE synthesis in both organs. However, penicillin-G sodium (0.25-1g kg-1 day-1 for 4 days i. p.) inhibited 12-HETE synthesis in both organs. Drug-induced changes in 15-HETE synthesis are probably mediated via direct effects on the phosphoiipase and lipoxygenase enzymes involved in its synthesis. These changes in 12-HETE synthesis may underly some of the biochemical mechanisms involved in the drugs-induced actions in some mammalian organs.
